I have a male and a female guinea pig (Willy and Flossy). One of them keeps making a rumbling noise. It nearly sounds like a chirp but it's deep down in their throat. Is this maybe a 'purring' sound? I just want to know if it means they're happy or annoyed or breeding etc.

i think it's a sound of dominance....or they dont like each other... its like when they make those noises, it means they dont like what is happening to them at that moment. i think so because there was a site where they provide sounds of guinea pigs and explanations of what they are.
 
is it just a noise, or does their whole body start to rumble?
 
It is called rumble-strutting. Usually done by males either b/c they are feeling frisky, or they are proving dominance to another male.

It's not a bad thing. However, is one of the pigs fixed so they cannot breed? If not you should separate them until the male can be neutered. Not only because of overpopulation issues, but also because if your female is older than 9 mos and has never had a litter her pelvic bones could be fusing together. That equals dead or very sick female pig and the piglets!!! (Or are they called pups? I can't remember! LOL)

The rumble is fine.
